Résumé

Résumé Homo sapiens et les Néandertaliens ont subi une hybridation au cours du Paléolithique moyen/supérieur, aboutissant au maintien de petites quantités d'ADN d'origine néandertalienne dans le génome humain moderne. Dans la présente étude, nous examinons les rôles potentiels que les polymorphismes mononucléotidiques (SNP) néandertaliens pourraient jouer dans la susceptibilité à l'autisme chez des personnes noires non hispaniques, blanches hispaniques et blanches non hispaniques, à l'aide de données issues des bases de données Simons Foundation Powering Autism Research (SPARK), Genotype-Tissue Expression (GTEx) et 1000 Genomes (1000G). Nous avons découvert que les variants rares sont significativement enrichis chez les probands autistes par rapport à des témoins appariés selon l'origine raciale. En outre, nous avons identifié 25 SNP rares et communs qui sont significativement enrichis dans l'autisme selon différents contextes ethniques, dont certains présentent des associations cliniques significatives. Nous avons également identifié d'autres SNP qui partagent des corrélations génotype-phénotype plus spécifiques mais qui ne sont pas nécessairement enrichis dans l'autisme et qui peuvent néanmoins jouer un rôle dans l'expression de phénotypes comorbides (p. ex., déficience intellectuelle, épilepsie et régression du langage). Ces résultats suggèrent fortement que l'ADN d'origine néandertalienne joue un rôle significatif dans la susceptibilité à l'autisme au sein des principales populations des États-Unis.

Abstract Homo sapiens and Neanderthals underwent hybridization during the Middle/Upper Paleolithic age, culminating in retention of small amounts of Neanderthal-derived DNA in the modern human genome. In the current study, we address the potential roles Neanderthal single nucleotide polymorphisms (SNP) may be playing in autism susceptibility in samples of black non-Hispanic, white Hispanic, and white non-Hispanic people using data from the Simons Foundation Powering Autism Research (SPARK), Genotype-Tissue Expression (GTEx), and 1000 Genomes (1000G) databases. We have discovered that rare variants are significantly enriched in autistic probands compared to race-matched controls. In addition, we have identified 25 rare and common SNPs that are significantly enriched in autism on different ethnic backgrounds, some of which show significant clinical associations. We have also identified other SNPs that share more specific genotype-phenotype correlations but which are not necessarily enriched in autism and yet may nevertheless play roles in comorbid phenotype expression (e.g., intellectual disability, epilepsy, and language regression). These results strongly suggest Neanderthal-derived DNA is playing a significant role in autism susceptibility across major populations in the United States.
