Résumé

RÉSUMÉ Homo sapiens et les Néandertaliens ont subi une hybridation au cours du Paléolithique moyen/supérieur, aboutissant à la conservation de faibles quantités d'ADN dérivé de Néandertal dans le génome humain moderne. Dans la présente étude, nous examinons les rôles potentiels que les polymorphismes nucléotidiques simples (SNP) néandertaliens géniques peuvent jouer dans la susceptibilité à l'autisme à l'aide de données des bases de données Simons Foundation Powering Autism Research (SPARK) et Genotype-Tissue Expression (GTEx). Nous avons découvert que les variants rares et peu fréquents sont significativement enrichis à la fois chez les proposants autistes euro-américains et afro-américains et chez leurs frères et sœurs non atteints, comparativement à des témoins appariés selon l'origine raciale. De plus, nous avons identifié 51 SNP (p51) significativement enrichis chez les cas d'autisme euro-américains, dont 13 se situent dans des gènes associés à l'autisme, ainsi que 1 SNP chez les proposants afro-américains. De plus, les SNP au sein du réseau p51 présentent un déséquilibre de liaison significatif les uns avec les autres, ce qui indique qu'ils peuvent plus souvent être cohérités dans l'autisme. Ces résultats suggèrent fortement que l'ADN dérivé de Néandertal joue un rôle significatif dans la susceptibilité à l'autisme au sein des principales populations des États-Unis.

ABSTRACT Homo sapiens and Neanderthals underwent hybridization during the Middle/Upper Paleolithic age, culminating in retention of small amounts of Neanderthal-derived DNA in the modern human genome. In the current study, we address the potential roles genic Neanderthal single nucleotide polymorphisms (SNP) may be playing in autism susceptibility using data from the Simons Foundation Powering Autism Research (SPARK) and Genotype-Tissue Expression (GTEx) databases. We have discovered that rare and uncommon variants are significantly enriched in both European- and African-American autistic probands and their unaffected siblings compared to race-matched controls. In addition, we have identified 51 SNPs (p51) significantly enriched in European-American cases of autism, 13 of which fall within autism-associated genes, as well as 1 SNP in African-American probands. In addition, SNPs within the p51 network display significant linkage disequilibrium with one another, indicating they may more often be co-inherited in autism. These results strongly suggest Neanderthal-derived DNA is playing a significant role in autism susceptibility across major populations in the United States.
