Résumé

L'autisme est un trouble du neurodéveloppement se manifestant par une altération des interactions sociales, des déficits des compétences de communication, des intérêts restreints et des comportements répétitifs. Dans les troubles du neurodéveloppement, neurodégénératifs et psychiatriques, les cellules gliales subissent des réarrangements morphologiques, biochimiques et fonctionnels, qui sont essentiels au développement neuronal, à la neurotransmission et à la connectivité synaptique. La fonction cérébelleuse ne se limite pas à la coordination motrice, mais contribue également à la cognition et peut être affectée dans l'autisme. Les oligodendrocytes et, plus particulièrement, les précurseurs oligodendrogliaux sont très sensibles au stress oxydatif et aux agressions excitotoxiques. Dans la présente étude, nous avons recherché des éléments en faveur d'une oligodendropathie développementale dans le contexte de l'autisme en réalisant une analyse en réseau de l'expression génique de tissu cérébelleux. Nous avons créé un modèle de réseau in silico (OLIGO) montrant le paysage des interactions entre les marqueurs oligodendrocytaires et démontré que plus de 50 % (16 sur 30) des gènes au sein de ce modèle présentaient des modifications significatives d'expression (valeur de p corrigée < 0,05) dans le cervelet de patients autistes. En particulier, nous avons observé une augmentation de l'expression des marqueurs oligodendrocytaires spécifiques de OLIG2, MBP, OLIG1 et MAG. Nous postulons qu'une expression aberrante de gènes spécifiques des oligodendrocytes, potentiellement liée à des modifications de l'oligodendrogenèse, pourrait contribuer à un développement cérébelleux anormal, à une myélinisation altérée et à une connectivité synaptique anormale dans les troubles du spectre de l'autisme (TSA).

Autism is a neurodevelopmental disorder manifested by impaired social interaction, deficits in communication skills, restricted interests, and repetitive behaviors. In neurodevelopmental, neurodegenerative, and psychiatric disorders, glial cells undergo morphological, biochemical, and functional rearrangements, which are critical for neuronal development, neurotransmission, and synaptic connectivity. Cerebellar function is not limited to motor coordination but also contributes to cognition and may be affected in autism. Oligodendrocytes and specifically oligodendroglial precursors are highly susceptible to oxidative stress and excitotoxic insult. In the present study, we searched for evidence for developmental oligodendropathy in the context of autism by performing a network analysis of gene expression of cerebellar tissue. We created an in silico network model (OLIGO) showing the landscape of interactions between oligodendrocyte markers and demonstrated that more than 50 % (16 out of 30) of the genes within this model displayed significant changes of expression (corrected p value <0.05) in the cerebellum of autistic patients. In particular, we found up-regulation of OLIG2-, MBP-, OLIG1-, and MAG-specific oligodendrocyte markers. We postulate that aberrant expression of oligodendrocyte-specific genes, potentially related to changes in oligodendrogenesis, may contribute to abnormal cerebellar development, impaired myelination, and anomalous synaptic connectivity in autism spectrum disorders (ASD).
